Come now, a vise? No, the fingers are used to gently feel the outer surfaces of the testicles. A vise has no tactile sense, for goodness sake! The doctor will wear examining gloves for cleanliness sake, but that's all. The gloves are thin and flexible enough to allow the doctor's fingers to feel all that they need to feel.

No, they use their hands. However, perhaps the "vice" you're talking about was actually an instrument to measure the size of the lump accurately?
